practical summary of microb 2



kiri : Bacitracin test (transparent colony-B complete haemolysis)
sensitive: Streptococcus pyogene
resistant: other than Group A

kanan : Optochin test 
(grennish discolouration-alpha partial heamolysis)

resistance : Streptococci Viridans 


kiri : mannitol salt agar (MSA)-selective media
kanan : Staphylococci Aureus
-ferment mannitol produce colonies 
surrounded by yellow halloes-

COLONY ON BLOOD AGAR : ada 3
1. alpha haemolytic streptococci
2. beta heamolytic streptococci
3. beta haemolytic staphyloccoci

alpha haemolytic streptocci 
(greenish discolouration)

perhatikan 2 perbezaan di bawah ini!!



beta haemolytic streptoccoci
(translucent pin point no pigmented colony surrounded by large haemolysis)



 beta haemolytic staphylococci
(white large pigmented colony 
surrounded by small zone of haemolysis)


2nd point to be noticed : 

staph - non fastidious (both nutrient & blood agar)
strep - fastidious (only in blood agar)

then, please recognize 4 things yang guna nutrient agar as medium:-

1. pigmented colony (kat atas)

2. Novobiocin test


resistance : Staphylococcus Saprophyticus
sensitive : Staphylococcus Epidermidis

3. antibiotic sensitivity test
disc diffusion method (staphylococci)



4.Phage typing


best method for typing Staphylococci


last test : HEAT RESISTANCE TEST
(Enterococcus fecalis)

E. faecalis can withstand heat at 60C for 30 mins

last but not least

blood culture media : DIPHASIC MEDIUM


slope of nutrient agar+10ml sulphonated broth

can commonly occur in arteries at the base of the brain (the circle of Willis) and an aortic aneurysm occurs in the main artery carrying blood from the left ventricle of the heart. When the size of an aneurysm increases, there is a significant risk of rupture, resulting in severe hemorrhage, other complications or death. Aneurysms can be hereditary or caused by disease, both of which cause the wall of the blood vessel to weaken

Cardiac tamponade, also known as pericardial tamponade, is an emergency condition in which fluid accumulates in the pericardium (the sac in which the heart is enclosed). If the fluid significantly elevates the pressure on the heart it will prevent the heart's ventricles from filling properly. This in turn leads to a low stroke volume. The end result is ineffective pumping of blood,shock, and often death.
Cardiac tamponade occurs when the pericardial space fills up with fluid faster than the pericardial sac can stretch. If the amount of fluid increases slowly (such as in hypothyroidism) the pericardial sac can expand to contain a liter or more of fluid prior to tamponade occurring. If the fluid occurs rapidly (as may occur after trauma or myocardial rupture) as little as 100 ml can cause tamponade.
